It should be taken into consideration that pancreas  secretes not only hormones, such as insulin, but also digestive enzymes, such as trypsin, chymotrypsin, and other polypeptidases. Grinding up the pancreas results in releasing of both insulin and digestive enzymes. But, digestive enzymes are able to break down insulin into fragment not anymore considered as a hormone. So, it is not possible to isolate insulin from this homogenate
